Tyrone is an anthropomorphic moose and the tritagonist of The Backyardigans. Tyrone's speaking voice was initially provided by Reginald Davis, with Jordan Coleman assuming the role during the second season. Chris Grant voiced the character for the final season. Tyrone's singing voice was first provided by Corwin Tuggles. Leon Thomas provided it in the second season and some of the third, with Damani Roberts taking over for several episodes and Tyrel Williams assuming the role for the remainder of the third and fourth seasons.

Uniqua is the protagonist of The Backyardigans. She is the only character who appears in every episode. She is a one-of-a-kind, unique creature, hence her name. The series uses the name "Uniqua" for both the character and her species. In an interview, the show's creator Janice Burgess said that Uniqua was based on herself: "Uniqua is me. Or at least who I was as a kid. She's a ringleader. Out of the five, Uniqua is the main character, and we use her in every story." Uniqua is voiced by LaShawn Tináh Jefferies throughout the entire series. Her singing voice is provided by Jamia Simone Nash in seasons one through three and by Avion Baker in the fourth season.

Pablo is a blue penguin and the deuteragonist of The Backyardigans. He does not appear in "Chichen-Itza Pizza," making him the character with the second-most appearances (after Uniqua, who appears in every episode). Pablo's voice was provided by Zach Tyler Eisen in the first season and by Jake Goldberg for the remainder of the series. His singing voice is provided by Sean Curley.

Barney is a purple dinosaur and the main protagonist of the television series Barney & Friends and its home video predecessor Barney & The Backyard Gang. A two hundred million year (two dinosaur years) old, six-foot tall purple tyrannosaurus-rex with a green tummy and green spots on his back and his tail and yellow toes, he comes to life through a child's imagination, springing up from a doll to full size. In turn, Barney opens children's eyes to the joys of learning and discovery, to the wonders of make-believe, and to the beauty of unconditional love. Dubbed as the "Elvis for Toddlers" in the early days, he is best known for his silly and optimistic attitude. His birthplace is Dallas, Texas (as he was created there by the creators), however, he lives and resides in one's imagination. His place of residence would later be expanded in the video Come on Over to Barney's House (2000). Although Barney comes to life through a child's imagination, he isn't imaginary. Once Barney springs to life from imagination, any and everyone can see him and no one else needs to use their imagination to see or interact with him in his large form. Created by former teacher Sheryl Stamps Leach, Barney first appeared in the video titled The Backyard Show (1988), which was the first episode of the Barney & The Backyard Gang home video series. David Voss originally performed as the costume for Barney from 1988 to 1990, and was succeeded by David Joyner, who would later define the costume mannerisms of the character. The character has been succeeded by many other costume performers during Joyner's time and after his resignation. The Barney character was originally voiced by Bob West from 1988 until 2000, but since his resignation has been voiced by many other voice actors. Barney generally appears with his dino pals, Baby Bop, BJ and or Riff among others. In 2023, after the series Barney's World was announced, franchise owner Mattel would split the brand into two different pillars for licensing purposes. To separate the long-known iteration from the iteration they created for the new series, this version of the character was dubbed "classic Barney"

Simon Seville is one of The Chipmunks and the overall deuteragonist of the series and films. He is the smartest of The Chipmunks, possessing an I.Q. just north of Einstein's. Simon possesses a very dry sense of humor, as well as a keen wit. Simon gets along great with Jeanette, who is smart, just like him. They are shy about their relationship, however it is obvious that he cares for her.

Alvin Seville is one of The Chipmunks and the overall main protagonist of the series and movies. He is the ringleader of the iconic trio. Alvin greatly relishes his role as band front man, and his impulsive behavior regularly gets him and his brothers (sometimes even Dave) into trouble. Despite all his faults, he is very kind, caring, friendly, sweet, good-hearted, loving and nice, and he has got a very big, gold heart.
